What Is Retatrutide?

Retatrutide (LY-3437943) is an investigational triple receptor agonist peptide that activates three metabolic receptors at once: GLP-1 (glucagon-like peptide 1), GIP (glucose-dependent insulinotropic polypeptide), and glucagon. The combined action of all three pathways is the basis for the compound being studied as one of the most potent investigational research compounds in this class.

In the 2023 Phase 2 trial published in *The New England Journal of Medicine*, the 12-mg retatrutide group had a least-squares mean body-weight change of −24.2% at 48 weeks, compared with −2.1% with placebo. In the completed Phase 3 TRIUMPH-1 trial, Lilly’s May 2026 topline results showed average weight changes at 80 weeks of −19.0% with 4mg, −25.9% with 9mg, and −28.3% with 12mg, compared with −2.2% with placebo. In a prespecified 104-week extension among participants with baseline BMI ≥35, the 12-mg group reached an average −30.3% weight change. How Retatrutide Works — Triple Receptor Mechanism

Retatrutide activates three receptor pathways simultaneously. No other metabolic compound described in the published literature does this.

GLP-1 (glucagon-like peptide 1) receptor

Promotes satiety signaling, slows gastric emptying, and stimulates glucose-dependent insulin secretion. The same pathway targeted by semaglutide and by the GLP-1 component of tirzepatide.

GIP (glucose-dependent insulinotropic polypeptide) receptor
